Inter-laboratory variation comes from real methodological differences, not sloppiness: which reference standard was used and how it was itself…

Thank you for spelling out the reasoning rather than just the conclusion.

From the other side of the consultation, briefly.

HPLC results interpretation for certificates of analysis: when you get your Janoshik report back, here's what the numbers mean:

  • Purity %: Should be >97%. Below 95% is concerning.
  • Retention time: Should match reference standard for the peptide
  • Related substances: Degradation products should be <2% total
  • Single impurity: No individual impurity should exceed 1%

My last batch: 99.6% purity, 0.2% total related substances. Clean chromatogram with single sharp peak at expected retention time. Exactly what you want to see.

Third-party testing services for certificates of analysis besides Janoshik: Janoshik: Gold standard for peptide community, ~$151, 10-14 day turnaround…

I tested my certificates of analysis batch through Janoshik — 97.6% purity. That's within acceptable range for compounded peptides. Turnaround was 8 business days.

The test costs ~$129 but gives you actual HPLC chromatography data plus mass spec confirmation. For the peace of mind alone, it's worth every penny. I test every new batch from a new vendor.

They are not contradictory, they measure different things, and this is the single most common misreading of a certificate.

Community-verified vendors for certificates of analysis: I maintain a spreadsheet of Janoshik-verified test results submitted by community members. Across 52 submitted tests:

  • 85% tested at ≥98% purity
  • 10% tested at 95-98% purity
  • 5% tested at <95% purity (concerning)

The vendors with consistently high purity results are listed in the Vendor Directory. We update monthly based on new community submissions.

Moderator note: leaving this open. It is being argued well and the disagreement is the useful part.
